The Octet® GlyS kit has been designed for a quick high-throughput relative Sialic acid content screening in both purified and crude cell culture samples during early clone selection stages in the CLD process. The kit can be used to screen and rank both mAb and non-mAb samples (e.g. high, medium or low) and when combined with titer analysis (which can be also performed on the Octet® platform), it facilitates the selection of the top performing cell lines producing proteins with the most favorable glycan (Sialic Acid) profile. The GlyS kit allows verified decisions to be made about which clones are the most suitable to move downstream for further optimization. Reliable glycosylation data (as a key part of Critical Quality Attribute (CQA) profile) at this early stage supports shorter CLD lead times and prevents later failures.
With the GlyS Kit, no sample purification or digestion is required. This allows for the analysis of crude samples. Now, you can screen to identify ideal candidates or failing candidates much faster and much earlier. The GlyS Kit can help reduce preparation time by up to three hours.
Simplified Assay Workflow
The Octet® GlyS Kit is based on the use of sialic acid specific lectin. It allows to analyze both crude and purified samples without the need for purification or enzymatic digestion. This essentially eliminates the need for any sample preparation beyond a dilution step, simplifying the sialic acid screening workflow for users. Additionally, the GlyS kit reduces time to results and increases analysis throughput by allowing to screen up to 96 clones in under one hour on the Octet® RH96 system.
Combine Sialylation and Titer Data on the Octet® Platform
Octet® Analysis Studio software enables effective combination of relative Sialic Acid content screening data and titer determination data (acquired on the Octet® platform) on the same sample. It allows verified decisions to be made about which clones are the most suitable to move downstream for further optimization.
